--------------------------------------------------------------------- COLLOQUIUM OF THE LABORATORY FOR COMPUTER DESIGN OF MATERIALS Institute for Computational Sciences and Informatics (CSI 898-Sec 001) Protein solution and crystallization dynamics: experimental and computational techniques Sridhar Gorti Naval Research Laboratory,Washington, DC Measurements of lysozyme diffusion within the "depletion zone" of the (110) face of lysozyme crystals are reported. The measurements were performed using the technique of microscope light scattering (MLS), capable of measuring small sampling areas (~2 µm2). Measurements within such small areas allowed for the determination of intensity correlation functions at precise locations (at ~10 µm intervals) adjacent to the (110) crystal face. Using well-established methods in the analyses of intensity correlation functions, the diffusion coefficients of lysozyme molecules adjacent to the (110) crystal face and in solution were obtained. The diffusion coefficients were observed to decrease as the surface of the crystal was approached. This decrease is shown to occur, in theory and model calculations, as a result of the depletion of lysozyme molecules, whose diffusion coefficients are ~8 x 10-7 cm2/s at 23 °C.
